Slice incubation chamber insert for electrophysiology.
A handy insert that can be used in combination with regular 200mL beakers to incubate slices for electrophysiology. To use, glue netting to the bottom of the holding part (either specialized slice netting or from netted cloth, e.g. pantyhose) and trim the flanges for a snug fit in your beaker. Medium versions should work with any beaker with an inner diameter between 54 to 64mm, while the "Large" version is designed for beakers of 82mm to 102mm inner diameter.
The central canal has a strut at the bottom to ensure your oxygen tube will not drop down too far, and intake holes to facilitate liquid flow. When oxygen is flowing, it will push up the liquid inside the canal, forcing a steady and even flow of oxyginated ACSF. Slice holding area is divided in 6 or 8 compartments, which are big enough to hold a sagital or coronal mouse brain slice for the medium sized inserts. Larger rat brains may not fit as well, though.
Performance so far
We currently use five "medium" sized inserts in our lab, to relative success. This replaced home-made solutions based on glueing bits of falcon tube together, and was a definite improvement. Note that some discoloration can occur, but after two years of use we have not seen any ill effects on our slices.
The large version remains untested so far, but should fit beakers around 500ml (depending on brand) and should be more than big enough for rat slices.
